چکیده
Introduction: Brucellosis is a chronic zoonotic disease caused by Brucella species that continues to pose major public health and veterinary challenges, particularly in endemic regions. While ruminants are traditionally recognized as the primary reservoirs, the role of domestic dogs, especially those in close contact with livestock, remains underexplored. This study investigated the presence of Brucella melitensis in indigenous dogs in Neyshabur and its surrounding rural areas using a combination of serological, bacteriological, and molecular diagnostic methods.
Materials & Methods: A total of 100 blood samples were collected from herding and stray dogs. Initial screening by the Rose Bengal plate test (RBPT) identified 31 seropositive samples. These samples were subjected to polymerase chain reaction (PCR) using both genus-specific (B4/B5) and species-specific (B. melitensis) primers.
Results: Only three samples (9.7% of RBPT-positive cases), all from herding dogs, were PCRpositive for B. melitensis, and none were positive among stray dogs. The isolated organisms were further characterized and confirmed as B. melitensis Biovar 1 through biotyping. No significant association was found between infection and sex, while a statistically significant relationship was observed between infection status and dog type (P=0.013), indicating a higher risk among herding dogs.
Conclusion: The results of this study suggest that dogs involved in herding activities may serve as epidemiological bridges between infected livestock and other animals or humans. The findings underscore the limitations of serological testing alone due to the possibility of false positives and support the inclusion of PCR as a critical tool for definitive diagnosis. Enhanced molecular surveillance of brucellosis in companion and working dogs is recommended for effective disease control and to minimize zoonotic risk in endemic regions like northeastern Iran.

تب مالت یک بیماری مزمن زئونوز است که توسط گونه‌های مختلف باکتری Brucella ایجاد می‌شود و همچنان چالشی جدی برای بهداشت عمومی و دامپزشکی، به‌ویژه در مناطق بومی محسوب می‌گردد. اگرچه نشخوارکنندگان به‌عنوان مخازن اصلی این بیماری شناخته می‌شوند، اما نقش سگ‌های خانگی، به‌ویژه آن‌هایی که در تماس نزدیک با دام قرار دارند، کمتر مورد بررسی قرار گرفته است. این مطالعه با هدف بررسی حضور Brucella melitensis در سگ‌های بومی شهرستان نیشابور و مناطق روستایی اطراف آن، از روش‌های تشخیصی سرولوژیک، باکتریولوژیک و مولکولی به‌صورت ترکیبی استفاده کرده است. مجموعاً ۱۰۰ نمونه خون از سگ‌های گله‌بان و ولگرد جمع‌آوری شد. در مرحله غربالگری اولیه با آزمون رزبنگال (RBPT)، ۳۱ نمونه سرواکسن مثبت شناسایی شد. این نمونه‌ها با استفاده از آغازگرهای اختصاصی جنس (B4/B5) و گونه (B. melitensis) تحت آزمایش PCR قرار گرفتند. تنها سه نمونه (۹/۷٪ از موارد مثبت RBPT) که همگی مربوط به سگ‌های گله‌بان بودند، در PCR برای Brucella melitensis مثبت شدند، در حالی که هیچ‌یک از سگ‌های ولگرد نتیجه مثبت نداشتند. ارگانیسم‌های جداشده از طریق بیوتیپینگ، به‌عنوان B. melitensis بیووار ۱ تأیید شدند. هیچ ارتباط معناداری بین ابتلا و جنسیت مشاهده نشد، اما ارتباط آماری معناداری بین وضعیت ابتلا و نوع سگ (p = 0.013) به‌دست آمد که نشان‌دهنده خطر بالاتر در میان سگ‌های گله‌بان است. این نتایج حاکی از آن است که سگ‌های فعال در امور گله‌داری ممکن است به‌عنوان پل‌های اپیدمیولوژیک میان دام‌های آلوده و سایر حیوانات یا انسان‌ها عمل کنند. یافته‌ها بر محدودیت‌های آزمون‌های سرولوژیک به‌تنهایی (به دلیل مثبت‌های کاذب) تأکید داشته و استفاده از PCR را به‌عنوان ابزاری کلیدی برای تشخیص قطعی پیشنهاد می‌کنند. پایش مولکولی تقویت‌شده در مورد تب مالت در سگ‌های همراه و کاری، برای کنترل مؤثر بیماری و کاهش خطر انتقال زئونوز در مناطقی مانند شمال شرق ایران توصیه می‌شود.
