چکیده
The Brucella bacterium, a facultative intracellular pathogen, is the causative agent of Brucellosis in humans and livestock. To overcome the limitations of traditional vaccines, contemporary research has increasingly focused on developing subunit vaccines. The protein sequences of BLS and L7/L12 were obtained from NCBI and the proposed epitopes of L7/L12 were obtained from the IEDB server. The most suitable L7/L12 epitopes, including the proposed B-cell and T-cell stimulatory epitopes that overlapped in some regions, were selected and inserted into non-conserved regions of the BLS protein. The VaxiJen server was used to assess the antigenicity of the sequence, and the SWISS-MODEL site was used to model the 3D structure of the recombinant protein. The Z-Score index from ProSA-web was used to evaluate the quality of the generated model, and the Ramachandran plot was generated using the Ramplot server. Other indicators, including the non-allergenicity and physicochemical properties such as stability and solubility, were also investigated. The antigenicity of the BLS sequence increased after insertion of the proposed epitopes, rising from 0.52 to 0.60 and 0.55 in models 1 and 2, respectively. The Z-Score of proposed structures 1 and 2 in ProSA-web were -6.6 and -5.57, respectively. According to the Ramachandran plot, no amino acids were observed in the disallowed region in either model, or any occurrences could be ignored. A check of the recombinant sequence in the IEDB server revealed the L7/L12 epitopes among the proposed epitopes. Other results confirmed the stability, appropriate solubility, and non-allergenicity of the recombinant protein.
طراحی و ارزیابی in silico کاندیداهای واکسن زیر واحدی مبتنی بر داربست BLS حاوی اپی توپهای مشتق شده از L7/L12 علیه بروسلوز

باکتری بروسلا، یک پاتوژن درون سلولی اختیاری، عامل ایجاد کننده بروسلوز در انسان و دام است.برای غلبه بر این محدودیتهای واکسن های سنتی، تحقیقات معاصر به طور فزایندهای بر توسعه واکسنهای زیر واحدی متمرکز شده است.هدف این مطالعه شناسایی اجزای ایمنیزای خاص باکتری بروسلا است که میتوانند پاسخ ایمنی قوی ایجاد کنند، در حالی که اجزای بیماریزای باکتری را از بین میبرند. توالی پروتئین BLS و L7/L12 از NCBI و اپیتوپهای پیشنهادی L7/L12 از سرور IEDB به دست آمد. مناسبترین اپیتوپهای L7/L12، از جمله اپیتوپهای تحریککننده سلولهای B و T که در برخی مناطق همپوشانی داشتند، انتخاب و در مناطق غیرحفاظتشده پروتئین BLS قرار داده شدند. از سرور VaxiJen برای ارزیابی آنتیژنیسیته توالی و از سایت SWISS-MODEL برای مدلسازی ساختار سهبعدی پروتئین نوترکیب استفاده شد. از شاخص Z-Score از ProSA-web برای ارزیابی کیفیت مدل تولید شده استفاده شد و نمودار Ramachandran با استفاده از سرور Ramplot ایجاد شد. سایر شاخصها، از جمله عدم حساسیتزایی و خواص فیزیکوشیمیایی مانند پایداری و حلالیت نیز بررسی شدند. آنتیژنیسیته توالی BLS پس ازدرج اپیتوپهای پیشنهادی افزایش یافت و به ترتیب از 0.52 به 0.60 و 0.55 در مدلهای 1 و 2 رسید. Z-Score ساختارهای پیشنهادی 1 و 2 در ProSA-web به ترتیب 6.6- و 5.57- بود. طبق نمودار Ramachandran، هیچ اسید آمینهای در ناحیه غیرمجاز در هیچ یک از مدلها مشاهده نشد، یا قابل چشمپوشی بود. بررسی توالی نوترکیب در سرور IEDB، اپیتوپهای L7/L12 را در میان اپیتوپهای پیشنهادی نشان داد. سایر نتایج، پایداری، حلالیت مناسب و غیرآلرژن بودن پروتئین نوترکیب را تأیید کرد.
