چکیده
Introduction: RSV is a single-stranded RNA virus with a nearly 15 kb genome containing 10 genes that encode 11 proteins. It has long been a leading cause of severe respiratory illness in children and the elderly. The G and F proteins are the most essential: G mediates attachment to host cells and stimulates immune responses, while both proteins directly contribute to the virus's pathogenicity. Objective: The aim of this study was to develop a potential antigenic fragment based on a conserved region of the RSV- G gene, which was selected due to its key role in virus attachment to host cells and triggering an immune response. Materials and Methods: For this purpose, five positive control RSV clinical samples were collected, and after RNA extraction, cDNA synthesis, and amplification of the central conserved region of the G gene using a nested PCR reaction, the resulting product was cloned into the pcDNA3.1+Hygro expression vector. Then, the transformation of Escherichia coli strain BL21 with the recombinant plasmid was performed, and after extraction, it was confirmed by enzymatic digestion and sequencing. Results: All molecular steps, also confirming their proper quality, cloning, and transformation, were completed successfully. Conclusion: The results showed that the central conserved region of the G gene was cloned into the target vector without unwanted mutations, and the next phases of vaccine design can proceed. This study is a preliminary step towards the development of an RSV candidate vaccine.
ساخت وکتور نوترکیب حاوی ژن زیر واحد G ویروس سنسیشیال تنفسی(RSV) به عنوان سازه مقدماتی واکسن

مقدمه: ویروس سنسیشیال تنفسی(RSV) یک ویروس RNA تکرشتهای با ژنومی تقریباً ۱۵ کیلوباز است که شامل ۱۰ ژن بوده و ۱۱ پروتئین را کد میکند. این ویروس مدتهاست که به عنوان یکی از عوامل اصلی بیماریهای شدید تنفسی در کودکان و سالمندان شناخته شده است. پروتئینهای G و F از مهمترین پروتئینهای این ویروس هستند؛ بهطوریکه پروتئین G نقش میانجی در اتصال ویروس به سلولهای میزبان و تحریک پاسخهای ایمنی را دارد، در حالی که هر دو پروتئین بهطور مستقیم در بیماریزایی ویروس نقش دارند.
هدف: هدف این مطالعه، توسعه یک قطعه آنتیژنی بالقوه بر اساس یک ناحیه حفاظتشده از ژن G ویروس RSV بود که به دلیل نقش کلیدی آن در اتصال ویروس به سلولهای میزبان و تحریک پاسخ ایمنی، انتخاب شد.
مواد و روشها: برای این منظور، پنج نمونه بالینی RSV مثبت به عنوان نمونههای کنترل جمعآوری شدند. پس از استخراج RNA، سنتز cDNA و تکثیر ناحیه حفاظتشده مرکزی (Central Conserved Region; CCR) ژن G با استفاده از واکنش Nested PCR، محصول حاصل در وکتور بیانی pcDNA3.1+Hygro کلون شد. سپس، ترانسفورماسیون سویه باکتری اشرشیا کولای سویه BL21 با پلاسمید نوترکیب انجام شد و پس از استخراج پلاسمید، سازه نوترکیب با استفاده از هضم آنزیمی و تعیین توالی تأیید گردید.
نتایج: تمام مراحل مولکولی، شامل تأیید کیفیت مناسب محصول، کلونینگ و ترانسفورماسیون، با موفقیت انجام شد.
نتیجهگیری: نتایج نشان داد که ناحیه حفاظتشده مرکزی ژن G بدون ایجاد جهشهای ناخواسته در وکتور هدف کلون شده است و مراحل بعدی طراحی واکسن میتوانند بر اساس این سازه ادامه یابند. این مطالعه یک گام مقدماتی در جهت توسعه یک کاندید واکسن RSV محسوب میشود.
