چکیده
Introduction: Tropical theileriosis caused by Theileria annulata is a major cattle disease in hyperendemic regions such as the Miankaleh Peninsula. This study evaluated molecular and serological diagnostic methods in vaccinated and non-vaccinated cattle. Materials and Methods: Blood samples from 28 vaccinated and 29 non-vaccinated cattle were analyzed using PCR targeting the Tams-1 gene and schizont antigen-based indirect ELISA (iELISA). Diagnostic performance and agreement between methods were assessed. Results: The iELISA showed high sensitivity but moderate specificity, likely due to cross-reactivity. PCR detected active T. annulata infection in 18/28 vaccinated cattle (64.3%) and 14/29 non-vaccinated cattle (48.3%), with no significant difference between groups (RR = 1.33; 95% CI: 0.82–2.16; P = 0.23). Antibodies were detected by iELISA in 23 vaccinated cattle (82.1%) and 21 non-vaccinated cattle (72.4%), with no significant difference between groups (RR = 1.13; 95% CI: 0.84–1.54; P = 0.40). Agreement between PCR and iELISA was moderate (κ = 0.55), with most discordant results being PCR-negative/iELISA-positive, consistent with previous exposure, vaccination, or antibody persistence. Dot-blot ELISA showed diagnostic performance comparable to iELISA while being simpler and more cost-effective. Conclusion: The in-house schizont-based iELISA, combined with PCR, is a useful approach for surveillance of T. annulata in endemic areas. Frequent PCR positivity in clinically healthy, including vaccinated, cattle indicates continued parasite circulation with subclinical infection, supporting the integration of vaccination with molecular surveillance.
رزیابی میدانی یک آزمون الایزای مبتنی بر آنتیژن شیزونتِ تهیهشده در آزمایشگاه (In-house ELISA) و واکنش زنجیرهای پلیمراز (PCR) برای شناسایی آنتی بادی ها و عفونت ناشی از Theileria annulata در گاوهای واکسینه و غیرواکسینه در شبهجزیره میانکاله، ایران

مقدمه: تیلریوزیس گرمسیری ناشی از Theileria annulata یکی از مهمترین بیماریهای گاو در مناطق بومی ایران مانند شبهجزیره میانکاله است. در این مطالعه، روشهای تشخیصی مولکولی و سرولوژیک در گاوهای واکسینه و غیرواکسینه مورد ارزیابی قرار گرفت.
مواد و روشها: نمونههای خون ۲۸ رأس گاو واکسینه و ۲۹ رأس گاو غیرواکسینه با استفاده از آزمون PCR مبتنی بر ژن Tams-1 و آزمون الایزای غیرمستقیم (iELISA) مبتنی بر آنتیژن شیزونت بررسی شدند. همچنین، عملکرد تشخیصی و میزان توافق بین دو روش ارزیابی شد.
یافتهها: آزمون iELISA حساسیت بالایی، اما ویژگی متوسطی نشان داد که احتمالاً ناشی از واکنشهای متقاطع بود. آزمون PCR عفونت فعال T. annulata را در ۱۸ مورد از ۲۸ گاو واکسینه (۶۴/۳ درصد) و ۱۴ مورد از ۲۹ گاو غیرواکسینه (۴۸/۳ درصد) شناسایی کرد و اختلاف معنیداری بین دو گروه مشاهده نشد (RR=1.33؛ فاصله اطمینان ۹۵٪: –2.16-0.82؛ P=0.23). آزمون iELISA آنتیبادی علیه T. annulata را در ۲۳ رأس گاو واکسینه (۸۲/۱ درصد) و ۲۱ رأس گاو غیرواکسینه (۷۲/۴ درصد) شناسایی کرد که این تفاوت نیز از نظر آماری معنیدار نبود (RR=1.13؛ فاصله اطمینان ۹۵٪: 1.54-0.84؛ P=0.40). میزان توافق بین PCR و iELISA در حد متوسط بود (κ=0.55) و بیشتر موارد عدم تطابق مربوط به نمونههای PCR منفی و iELISA مثبت بود که با مواجهه قبلی، واکسیناسیون یا پایداری آنتیبادیها سازگار است. آزمون داتبلات الایزا نیز عملکرد تشخیصی قابل مقایسهای با iELISA داشت، در حالی که از نظر اجرا سادهتر و از نظر هزینه مقرونبهصرفهتر بود.
نتیجه گیری: الایزای مبتنی بر آنتیژن شیزونت، در ترکیب با آزمون PCR، روشی سودمند برای پایش Theileria annulata در مناطق آندمیک است. فراوانی بالای نتایج مثبت PCR در گاوهای از نظر بالینی سالم، از جمله دامهای واکسینه، نشاندهنده تداوم گردش انگل همراه با عفونتهای تحتبالینی است و بر ضرورت تلفیق برنامههای واکسیناسیون با پایش مولکولی تأکید میکند.
