چکیده
سابقه و هدف: تغییر سریع سبک زندگی انسان، تقاضا برای مصرف مواد غذایی فرآوری شده و بسته بندی شده را افزایش داده است. ماهیت بازدارنده بستهبندی باعث کاهش قرارگرفتن اجزای داخلی غذا با محیط خارجی مانند میکروارگانیسمها، هوا و نور میشود و بنابراین فساد موادغذایی را به تأخیر میاندازد. امروزه فیلمهای خوراکی بر پایه پلیمرهای طبیعی بویژه پلی ساکاریدهای خوراکی به دلایل زیست محیطی، تجزیهپذیری و ایمن بودن مورد توجه قرار گرفتهاند. پوششهای خوراکی با ایجاد لایهی نازک روی مواد غذایی از آنها در برابر عوامل فساد محافظت کرده و عمر مفید مواد غذایی را افزایش میدهند. ضمن اینکه پوششهای خوراکی مشکلات آلودگی و زیست تخریبپذیری بستهبندیهای مشتق شده از منابع نفتی را مرتفع می سازند. هدف این پژوهش، بررسی تأثیر نانولولههای هالوسیت روی ویژگیهای فیلم خوراکی بر پایه موسیلاژ استخراج شده از بذر کتان است. مواد و روشها: در این پژوهش، ابتدا موسیلاژ بذر کتان با روش خیساندن در آب و رسوب دهی با اتانول استخراج شد. در مرحله بعد، پوشش-های خوراکی زیستتخریبپذیر بر پایه موسیلاژ بذرکتان با روش قالبریزی و تبخیر حلال تهیه شد. اثر درصدهای مختلف نانولوله هالوسیت (1، 3 و 5 درصد وزنی/وزنی) بر روی ویژگیهای جذب رطوبت، ممانعتی، نوری و حرارتی فیلمهای خوراکی موسیلاژ ارزیابی شد. برای شناسایی برهمکنشهای احتمالی میان پلیساکارید موسیلاژ بذر کتان و نانولولههای هالوسیت از آنالیز تبدیل فوریه مادون قرمز استفاده شد. پایداری حرارتی نمونهها با دستگاه توزین حرارتی در دامنه دمای اتاق تا ۶۰۰ درجه سلسیوس و تحت جریان نیتروژن بررسی شد. همچنین، پراکنش نانولولهها در ماتریس پلیمری با نقشهبرداری توزیع اتمهای سیلیس و آلومینیوم توسط طیفسنجی پراش انرژی پرتو ایکس ارزیابی شد. جذب رطوبت فیلمها در رطوبت نسبی ۷۵% از طریق وزنسنجی متوالی و نفوذپذیری بخار آب با استفاده از استاندارد ASTM E96 اندازهگیری شد. خواص مکانیکی فیلمها نیز مطابق استاندارد ASTM D882 بررسی شد. پارامترهای رنگی فیلمها شامل روشنایی، سبزی-سرخی و زردی-آبی با دستگاه رنگسنج ارزیابی شد. خواص ضدمیکروبی فیلمهای خوراکی در مقابل باکتریهای گرم منفی اشریشیاکلی و گرم مثبت استافیلوکوکوس اورئوس با روش شمارش کلونی بررسی شد. همچنین زیست تخریبپذیری فیلمها از طریق دفن در خاک به مدت 40 روز به صورت بصری مورد بررسی قرارگرفت. نتایج: نتایج FTIR نشان داد که شدت باند جذب مربوط به گروههای هیدروکسیل در cm-1 ۳۲۹۱ با افزودن نانولولههای هالوسیت کاهش یافته است که بیانگر تشکیل پیوندهای هیدروژنی میان گروههای هیدروکسیل موسیلاژ و سطوح نانولوله میباشد. افزایش میزان هالوسیت موجب بهبود پایداری حرارتی فیلمها شد؛ بهطوریکه فیلم حاوی 5% هالوسیت بیشترین مقاومت حرارتی را در میان نمونههای بررسیشده نشان داد. نقشهبرداری عنصری نیز افزایش تراکم نسبی سیگنالهای سیلیس و آلومینیوم را با افزایش مقدار هالوسیت نشان داد و بیشترین تراکم این عناصر در فیلم حاوی 5% هالوسیت مشاهده شد. جذب رطوبت فیلمها با افزایش مقدار هالوسیت از 3 به 5% بهطور معنیداری کاهش یافت و فیلم حاوی 5% هالوسیت با جذب رطوبت 54.8%، بهترین عملکرد ممانعتی در برابر رطوبت را نشان داد.، ضمن اینکه نفوذپذیری بخار آب نیز روند کاهشی مشابهی را دنبال کرد و از 5/11 برای فیلم کنترل به (g mm/(m^2 h kPa)) ۹/۵ برای فیلم ۵ % هالوسیت رسید. افزایش محتوی نانولوله موجب بهبود استحکام کششی فیلم موسیلاژ شد، بهطوریکه استحکام کششی فیلم حاوی 5% نانولوله به 31/69 مگاپاسکال رسید. پارامترهای رنگسنجی فیلمها با افزودن نانولوله هالوسیت تغییر کردند؛ بهطوریکه افزایش شاخصهای روشنایی (L*) و سفیدی و کاهش شاخص زردی (b*) مشاهده شد. فعالیت ضدباکتریایی فیلمها نیز با افزودن نانولوله هالوسیت به طور چشمگیری افزایش یافت. در نهایت، آزمون زیستتخریبپذیری با دفن در خاک به مدت ۴۰ روز نشان داد که فیلم موسیلاژ خالص، در مقایسه با فیلمهای حاوی نانولوله هالوسیت، تخریب ظاهری سریعتری داشت و با افزایش مقدار نانولوله هالوسیت، روند ظاهری تخریب فیلمها به تأخیر افتاد؛ بهطوریکه فیلم حاوی ۵ % هالوسیت کندترین روند ظاهری تخریب را در میان نمونهها نشان داد. نتیجه گیری: به کارگیری نانولوله هالوسیت به عنوان تقویت کننده میتواند ویژگیهای عملکردی و خواص فیلم خوراکی بر مبنای موسیلاژ بذرکتان را بهبود بخشد. در میان نمونههای مورد بررسی، فیلم حاوی ۵ درصد نانولوله هالوسیت بیشترین پایداری حرارتی و کمترین جذب رطوبت را نشان داد و همچنین از استحکام کششی و فعالیت ضدباکتریایی قابلقبولی برخوردار بود. بنابراین، فیلمهای موسیلاژ بذر کتان تقویتشده با نانولولههای هالوسیت، بهویژه در غلظت ۵%، دارای پتانسیل استفاده در توسعه بستهبندیهای خوراکی زیستتخریبپذیر هستند.

Background and objectives: Rapid changes in lifestyle have increased the demand for processed and packaged foods. The barrier properties of packaging limit the exposure of food components to external factors, such as microorganisms, air, and light, thereby delaying food spoilage. In recent years, edible films based on natural polymers, particularly polysaccharides, have attracted considerable attention owing to their biodegradability, safety, and environmental advantages. Edible coatings form a thin protective layer on food products, helping to protect them from spoilage factors and extend their shelf life. They also offer a potential alternative to petroleum-based packaging materials and may help mitigate their associated environmental concerns. Therefore, this study aimed to investigate the effects of halloysite nanotubes (HNTs) on the properties of an edible film based on flaxseed mucilage (FM). Methodology: Flaxseed mucilage was extracted by water soaking followed by ethanol precipitation. Biodegradable edible films based on FM were subsequently prepared using the solvent-casting and evaporation method. The effects of different HNT concentrations (1, 3, and 5 wt%) on the moisture absorption, barrier, optical, and thermal properties of the FM films were evaluated. Fourier-transform infrared (FTIR) spectroscopy was used to investigate potential interactions between FM and HNTs. The thermal stability of the films was evaluated by thermogravimetric analysis (TGA) from room temperature to 600 °C under a nitrogen atmosphere. The distribution of HNTs within the polymer matrix was qualitatively assessed by elemental mapping of Si and Al using energy-dispersive X-ray spectroscopy (EDX). Moisture absorption was determined by sequential weighing at 75% relative humidity, while water vapor permeability (WVP) was measured according to ASTM E96. The mechanical properties were evaluated according to ASTM D882. Film color parameters, including lightness (L*), redness–greenness (a*), and yellowness–blueness (b*), were determined using a colorimeter. The antibacterial activity of the films against the Gram-negative bacterium Escherichia coli and the Gram-positive bacterium Staphylococcus aureus was evaluated using the colony-counting method. Film biodegradability was also visually assessed following soil burial for 40 days. Results: FTIR analysis showed a decrease in the intensity of the hydroxyl-associated absorption band at 3291 cm⁻¹ following HNT incorporation, suggesting possible hydrogen-bonding interactions between the hydroxyl groups of FM and the HNT surfaces. Thermal stability increased with increasing HNT content, with the 5% HNT-containing film exhibiting the highest thermal stability among the formulations investigated. EDX mapping showed an increase in the relative density of Si and Al signals with increasing HNT content, with the highest signal density observed in the film containing 5% HNT. Moisture absorption significantly decreased as the HNT content increased from 3% to 5%; among the formulations investigated, the 5% HNT-containing film exhibited the lowest moisture absorption (54.8%). WVP showed a similar decreasing trend, decreasing from 11.5 for the control film to 5.9 for the film containing 5% HNT. Increasing HNT content also improved the tensile strength of the FM films, with the 5% HNT-containing film reaching a tensile strength of 69.31 MPa. HNT incorporation altered the color characteristics of the films, as indicated by increased lightness (L*) and whiteness and decreased yellowness (b*). The antibacterial activity of the films also increased markedly following HNT incorporation. After 40 days of soil burial, the pure FM film exhibited faster apparent degradation than the HNT-containing films, while increasing HNT content delayed the apparent degradation process; the 5% HNT-containing film exhibited the slowest apparent degradation among the formulations investigated. Conclusion: Overall, the incorporation of HNTs as a reinforcing agent improved several functional and physicochemical properties of FM-based edible films. Among the formulations investigated, the film containing 5% HNT exhibited the highest thermal stability and lowest moisture absorption, together with improved tensile strength and antibacterial activity. These findings suggest that HNT-reinforced FM films, particularly those containing 5% HNT, have potential for use in the development of biodegradable edible food-packaging materials.
