چکیده
سابقه و هدف: در طبیعت، گیاهان در معرض تنش های زیستی و غیر زیستی متعددی قرار می گیرند. با افزایش ارتفاع تغییرات دما، شدت تابش نور و رطوبت رشد گیاهان را محدود می کنند. مهمترین عاملی که در ارتفاعات باعث محدودیت رشد می شود، افزایش شدت تابش نور می باشد. در این شرایط، گیاهان برای جلوگیری از اثرات مضر شدتهای بالاتر تابش نور، سازگاریهای مورفوفیزیولوژیکی و بیوشیمیایی متنوعی ایجاد کردهاند که به عنوان مکانیسمهای حفاظت نوری تعریف میشوند. در این مطالعه برای اولین بار، نقش تنش نور زیاد و UVB بر تنظیم مکانیسمهای حفاظت نوری در دو اکوتیپ گیاه دارویی پونهسای البرزی (Nepeta crassifolia Boiss. & Buhse) بررسی شد. در حقیقت، تابش UVB به میزان معینی بر گیاهان دارویی، می تواند راهی مناسب برای بدست آوردن مقدار قابل توجهی از متابولیتهای ثانویه و در نتیجه افزایش ارزش های دارویی و اقتصادی آنها، باشد. مواد و روش ها: سه تا پنج بذر پونهسای البرزی از هر یک از اکوتیپ های ارتفاع پایین (2400 متری) و ارتفاع بالا (3500 متری) در گلدانهای پلاستیکی به عمق 40 سانتی متر و قطر 30 سانتی متر و حاوی خاک ماسه ای پرلیت دار به مدت 12 هفته کشت شدند و هر 7 روز یک بار با آب مقطر آبیاری شدند. گیاهان به شرایط نرمال اتاق رشد شامل فتوپریود 14 ساعت روشنائی و 10 ساعت تاریکی و دمای روز 25 و شب 18 درجه سانتی گراد منتقل شدند. گیاهان انتخاب شده به مدت 10 روز با شدت نور μmol m-2 s-1 400 و μmol m-2 s-1 800 و همچنین با پرتوفرابنفش با دوز 15 و 30 کیلوژول بر متر مربع در روز تیمار شدند. سنجش پارامترهای فلوئورسانس کلروفیل بر روی سومین برگ جوان و قبل از برداشت و توزین گیاهان انجام شد و سنجش رنگیزهها و متابولیتها بر روی نمونههای تازه برداشت شده و یا نگهداری شده در ازت مایع انجام شد. نتایج: هنگامی که اکوتیپهای مناطق مرتفع (ارتفاع 3500 متری) در معرض تنش ترکیبی (UVB+HL) قرار گرفتند، سازوکارهای حفاظت نوری بهتری در مقایسه با گیاهان ساکن ارتفاع پایین (ارتفاع 2400 متری) نشان دادند. گیاهان ساکن ارتفاع پایین، کاهش کلروفیل و فعالیت فتوشیمیایی را بروز دادند که منجر به وقوع مهار نوری فتوسیستمها شد. وقوع مهار نوری در این گیاهان با کاهش بیشینه عملکرد کوانتومی فتوسیستم II (Fv/Fm) و فعالیت کمپلکس فتولیز کننده آب در فتوسیستمII (Fv/Fo) همراه بود. مقاومت بالای اکوتیپهای مناطق مرتفع در معرض تنش ترکیبی افزایش سازگاری با شرایط نور بالا از طریق تجمع متابولیتهای آنتی اکسیدانی شامل ترکیبات فنلیک و کاروتنوئیدها انجام گرفت و در نتیجه عملکرد فتوشیمیایی برگها افزایش یافت. نتیجه گیری: به طور کلی تحت تنش ترکیبی در اکوتیپهای مرتفع مقادیر فنل کل، فعالیت آنزیم PAL، اسید رزمارینیک و اسید کلروژنیک به طور قابل توجهی بیشتر از اکوتیپهای کم ارتفاع بود. افزایش مقادیر پارامترهای فوق باعث بهبود فعالیت فتوشیمیایی فتوسنتز و در نتیجه افزایش تحمل اکوتیپهای مرتفع به تنش ترکیبی در مقایسه با اکوتیپهای ساکن ارتفاع پایین شد. یافتههای حاضر اطلاعات مهمی برای سازوکارهای دخیل در حفاظت نوری گیاه پونهسای البرزی در برابر تنشهای نوری ارائه می کند که آسیب به گیاهان را به حداقل رسانده و منجر به انتخاب اکوتیپهای مقاوم به شرایط تنش با ترکیبات مؤثره بیشتر به منظور استفادههای علمی، کشاورزی و صنعتی میشود.
Photoprotection mechanisms in Nepeta crassifolia ecotypes under UVB radiation and high light stress

Background and objectives: In nature, plants are continually exposed to abiotic and biotic stresses. Under altitudinal gradients, the environmental change in temperature, radiation, and moisture negatively affects plant growth. Among all the environmental factors that change with the altitude, high radiation is the most important factors limiting the plant growth. In such a condition, plants have developed various morphophysiological and biochemical adaptations, which are defined as photoprotection mechanisms, to avoid the harmful effects of higher intensities of light radiation. In this study, for the first time the role of high light stress and UVB on the regulation of photoprotection mechanisms in two ecotypes of Nepeta crassifolia plants was investigated. Indeed, a certain amount of UVB radiation on medicinal plants can be a suitable way to obtain a significant content of secondary metabolites and thus increase their medicinal and economic values. Methodology: Three to five seeds of Nepeta crassifolia were transferred and sown on the surface of the cylindrical plastic pots (30 cm in diameter and 40 cm in depth) containing sandy soil mixed with peat moss and perlite for 12 weeks, and were irrigated with distilled water every 7 days. Plants were grown at 25 °C day/14 °C night temperatures, with 16 h photoperiods and a daytime photon flux density of 400 μmol m–2 s–1, in an environmentally controlled growth chamber for a period of three months prior to the start of experiments. The mature plants were then exposed to irradiation with UVB (control, UVB, 15 and 30 kJ m-2 d-1) and high-intensity light (HL, 800 µmol m-2 s-1) stress for further 10 days. Measurements of chlorophyll fluorescence (which were described in the following section) and other analysis were performed on the fully expanded and mature leaves. For the latter analysis, leaf samples were freezed immediately in liquid N2 and stored until assay. Results: After exposure to UVB and HL stress, high altitude (3500 m) plants exhibited more tolerance to photoinhibition as compared to low-altitude plants. Under stress combination, low-altitude (2400 m) plants exhibited lower photochemical activity, which was associated with the down-regulation of chlorophyll, and consequently the occurrence of photoinhibition. The photoinhibition of PSII under combined stress was assessed by the analysis of maximal quantum yield of photosystem II (Fv/Fm), which was consistent with the decrease in the activity of oxygen-evolving complex (Fv/Fo). Increased tolerance in high altitude plants was accomplished by higher levels of phenol content under UVB and HL stress coupled with higher levels of carotenoids, and consequently higher photochemical functioning. Conclusion: To sum up, exposure of high altitude plants to HL and UVB radiation caused further increases in the activity of phenylalanine ammonia lyase (PAL) enzyme as well as the accumulation of phenolic, rosmarinic acid and chlorogenic acid (CGA), which play a key role in enhancing resistance to combined stress in comparison to low-altitude plants. The present findings provide important information about the selection of resistant ecotypes to stress conditions with more valuable compounds. The results of this study are useful for scientific, agricultural and industrial purposes.
