چکیده
مقدمه: ویروس بیماری نیوکاسل (NDV) یک پارامیکسوویروس پرندگان است که پرندگان اهلی و وحشی را تحت تأثیر قرار میدهد و بسته به سویه ویروسی، علائمی از شدید تا بدون علامت ایجاد میکند. در سالهای اخیر، چندین سویه از این ویروس به دلیل ویژگیهای انکولیتیک خود توجه زیادی را برای کاربردهای بالینی جلب کردهاند. بی خطری نسبی این ویروس در انسان، آن را به یک عامل ویروتراپی امیدوارکننده برای درمان بدخیمیهای انسانی تبدیل کرده است. بسته به سویه ویروس، سویههای کمحدت بهعنوان واکسن علیه بیماری نیوکاسل ، ایمن در نظر گرفته میشوند. به نظر میرسد که تمامی سویههای NDV توانایی تکثیر انتخابی در سلولهای توموری موش و انسان را دارند. با این حال، سویههای لنتوژنیک رفتار تکثیر تکچرخهای نشان میدهند، در حالی که سویههای بیماریزاتر تکثیر چندچرخهای دارند. در حوزه انکولوژی، این سویهها برای استفاده در ویروتراپی انکولیتیک در پستانداران نیز مورد مطالعه قرار گرفتهاند. هدف: این مطالعه مروری به بررسی ژنتیک ویروس، مکانیسمهای فعالیت انکولیتیک، تفاوتهای رفتاری میان سویههای NDV در پاسخ به سلولهای سرطانی و ویژگیهای سویههای مورد استفاده در درمان سرطان و چالشهای مرتبط با تولید آنها میپردازد. یافتهها: چندین سویه با ویژگیهای متفاوت مانند La Sota، MTH-68/H و NDV-HUJ بهعنوان سویههای انکولیتیک مورد استفاده قرار میگیرند. و سویه هایی نیز با مهندسی ژنتیک، تغییر یافته تا دردرمان سرطان اثربخشی بیشتری را نشان دهند. نتیجهگیری: به دلیل ایمنیزایی بالای NDV در انسان، نبود ایمنی پیشین، اختصاصیت نسبت به سلولهای سرطانی و توانایی تحریک سیستم ایمنی، این ویروس بهعنوان یک گزینه بسیار امیدوارکننده برای درمان سرطان مطرح است. برای تولید انبوه سویههای انکولیتیک و با توجه به محدودیتهای روشهای سنتی تکثیر مبتنی بر تخممرغ SPF، توسعه روشهای کشت سلولی و شناسایی ردههای سلولی جدید پیشنهاد میشود.

Introduction: Newcastle disease virus (NDV) is an avian paramyxovirus that affects both domestic and wild birds, causing symptoms that range from severe to asymptomatic depending on the viral strain. Several NDV strains have attracted considerable interest in recent years for clinical applications due to their oncolytic properties. Its safety in humans makes NDV a promising virotherapeutic agent for the treatment of human malignancies. Depending on the viral strain, less virulent strains are considered safe for use as vaccines against Newcastle disease. It appears that all NDV strains exhibit tumor-selective replication in both mouse and human cells. However, lentogenic strains display monocyclic replication behavior, whereas more pathogenic strains show multicyclic replication. In the field of oncology, these strains are also being studied for use in oncolytic virotherapy in mammals. Objective: This review study investigates the genetics of the virus, the mechanisms of oncolytic activity, the behavioral differences among NDV strains in their response to cancer cells , the characteristics of oncolytic NDV strains used in cancer therapy, and the challenges associated with their production. Findings: Multiple strains with different characteristics, such as La Sota, MTH-68/H, and NDV-HUJ, are used as oncolytic strains. Some strains have also been genetically engineered to show greater effectiveness in cancer treatment. Conclusion: Due to its high immunogenicity in humans, lack of pre-existing immunity, specificity for cancer cells, and ability to stimulate the immune system, NDV is considered a highly promising candidate for cancer therapy. For the mass production of oncolytic strains, and considering the limitations of traditional SPF egg-based production systems, the development of cell culture methods and the identification of new cell lines are proposed
