| Introduction: Cancer remains a major global health challenge, with high rates of morbidity and mortality. Traditional cancer treatments often cause adverse side effects due to their non-specific nature, leading to the exploration of targeted therapies, such as immunotoxins. Despite their potential, these therapies face hurdles, including the immunogenicity, off-target toxicity and low effectiveness. This research aims to explore a novel immunotoxin with reduced immunogenicity, improved safety and efficacy. Materials and Methods: Through a comprehensive bioinformatics analysis, this study mainly evaluates the immunogenicity and docking profile of the novel recombinant immunotoxin. The designed immunotoxin created by combining the shortest truncated form of diphtheria toxin (consisting of 380 amino acids) with a single-chain variable fragment monoclonal antibody synthesized in E. coli system for specifically targeting the human CD19 antigen. Results: In silico analyses show that the newly designed immunotoxin has acceptable immunogenicity and an improved targeting profile compared to existing versions and other B-cell malignancy immonotheraputics. This suggests its potential as a more effective and safer option for targeted therapy Conclusion: The findings declare DT380-CD19 immunotoxin potential, as a more effective and safer option for B-cell malignancy targeted therapy than former theraputics. The research underscores the necessity of integrating both computational and experimental approaches to refine immunosuppressive techniques without compromising the therapeutic effectiveness of immunotoxin therapies, ultimately striving to improve patient outcomes in cancer treatment. | ||

| مقدمه: سرطان همچنان یک چالش بزرگ سلامت جهانی با میزان بالای ابتلا و مرگ و میر است. درمانهای سنتی سرطان اغلب به دلیل عملکرد غیر اختصاصی خود عوارض جانبی نامطلوبی ایجاد میکنند که منجر به کشف درمانهای هدفمند مانند ایمونوتوکسینها شده است. با وجود پتانسیل بالای آنها، این درمانها با موانعی از جمله ایمنیزایی، عوارض جانبی و اثربخشی پایین مواجه هستند. هدف این تحقیق بررسی یک ایمونوتوکسین جدید با ایمنیزایی کاهشیافته، ایمنی و اثربخشی بهبود یافته است. مواد و روشها: این مطالعه از طریق یک تجزیه و تحلیل جامع بیوانفورماتیکی، عمدتاً ایمنیزایی و برهمکنش داکینگ ایمونوتوکسین نوترکیب جدید را ارزیابی میکند. ایمونوتوکسین طراحیشده با ترکیب کوتاهترین شکل کوتاهشده سم دیفتری (شامل ۳۸۰ اسید آمینه) با یک آنتیبادی مونوکلونال تکزنجیرهای با قطعه متغیر سنتز شده در سیستم بیان E. coli برای هدفگیری اختصاصی آنتیژن CD19 انسانی، ایجاد شده است. نتایج: آنالیزهای in silico نشان میدهد که ایمونوتوکسین جدید طراحی شده، ایمنیزایی قابل قبولی دارد و در مقایسه با نسخههای موجود ایمونوتراپی، پروفایل داکینگ بهبود یافتهای دارد. این موضوع، پتانسیل DT380-CD19 را به عنوان گزینهای مؤثرتر و ایمنتر برای درمان هدفمند بدخیمیهای سلولهای B CD19+ نشان میدهد. نتیجهگیری: یافتهها، پتانسیل ایمونوتوکسین DT380-CD19 را به عنوان گزینهای مؤثرتر و ایمنتر برای درمان هدفمند بدخیمی سلولهای B نسبت به روشهای درمانی قبلی اعلام میکنند. تحقیقات آینده باید بر ضرورت ادغام رویکردهای محاسباتی و تجربی برای اصلاح ایمنیزایی بدون به خطر انداختن اثربخشی درمان ایمونوتوکسین تأکید کند و در نهایت برای بهبود نتایج بیمار در درمان سرطان تلاش کند. | ||
