| Abstract The increasing prevalence of drug-resistant fungal infections, particularly oropharyngeal candidiasis (OPC), represents a major global health concern, especially among immunocompromised populations. This review critically evaluates the potential of actinomycete-derived secondary metabolites as alternative or adjunct therapeutic agents against antifungal-resistant fungal pathogens, with a primary focus on Candida albicans. Actinomycetes are well recognized for their capacity to produce structurally diverse bioactive compounds, including polyketides and non-ribosomal peptides, many of which exhibit potent antifungal activity and novel mechanisms of action capable of bypassing established resistance pathways. This review summarizes current knowledge on the antifungal efficacy of actinomycete metabolites, their mechanisms of action, and reported synergistic interactions with conventional antifungal agents. In addition, key limitations hindering clinical translation—such as cultivation challenges, toxicity concerns, formulation barriers, pharmacokinetic constraints, and regulatory considerations—are critically discussed. By integrating microbiological, pharmacological, and translational perspectives, this review highlights both the promise and the remaining gaps in the development of actinomycete-derived antifungal agents. Addressing these challenges through advanced genomic tools, rational screening strategies, and translational research is essential for advancing actinomycetes toward clinical application in OPC and other drug-resistant fungal infections. | ||

| افزایش شیوع عفونتهای قارچی مقاوم به دارو، بهویژه کاندیدیازیس اوروفارنژیال (OPC)، بهعنوان یک نگرانی عمده در بهداشت جهانی مطرح است، بهخصوص در جمعیتهای دچار نقص ایمنی. این مقاله مروری بهطور انتقادی پتانسیل متابولیتهای ثانویه مشتق از اکتینومیستها را بهعنوان عوامل درمانی جایگزین یا کمکی در برابر پاتوژنهای قارچی مقاوم به دارو، با تمرکز اصلی بر Candida albicans، مورد ارزیابی قرار میدهد. اکتینومیستها بهخوبی بهدلیل توانایی در تولید ترکیبات زیستفعال با تنوع ساختاری بالا، از جمله پلیکتیدها و پپتیدهای غیرریبوزومی، شناخته شدهاند که بسیاری از آنها دارای فعالیت ضدقارچی قوی و مکانیسمهای اثر نوینی هستند که قادر به دور زدن مسیرهای شناختهشده مقاومت میباشند. این مطالعه، دانش موجود درباره اثربخشی ضدقارچی متابولیتهای اکتینومیستی، مکانیسمهای عملکرد آنها، و تعاملات سینرژیستی گزارششده با داروهای ضدقارچی متداول را خلاصه میکند. علاوه بر این، محدودیتهای کلیدی که انتقال این ترکیبات به کاربردهای بالینی را با چالش مواجه میسازند—از جمله دشواریهای کشت، نگرانیهای سمیت، موانع فرمولاسیون، محدودیتهای فارماکوکینتیکی، و ملاحظات رگولاتوری—بهصورت انتقادی مورد بحث قرار میگیرند. با تلفیق دیدگاههای میکروبیولوژیک، فارماکولوژیک و پژوهشهای انتقالی (از آزمایشگاه به بالین)، این مقاله مروری هم ظرفیتهای بالقوه و هم شکافهای موجود در مسیر توسعه عوامل ضدقارچی مشتق از اکتینومیستها را برجسته میسازد. پرداختن به این چالشها از طریق بهکارگیری ابزارهای ژنومی پیشرفته، راهبردهای غربالگری هدفمند، و پژوهشهای انتقالی (از آزمایشگاه به بالین)، برای پیشبرد اکتینومیستها بهسوی کاربرد بالینی در درمان OPC و سایر عفونتهای قارچی مقاوم به دارو ضروری است. | ||
