چکیده
Abstract Introduction: Understanding Plasmodium infections in camels is of great importance for animal health and for assessing potential zoonotic transmission risks. Hence, the present study aims to explore the prevalence of Plasmodium infection in camels in the province of Sistan and Baluchestan in Iran. Material and Methods: A cross-sectional study was conducted from November 2022 to July 2024. A total of 270 camels (111 samples in spring, 93 in summer, 41 in autumn, and 25 in winter) were randomly selected from Sistan and Baluchestan province, and 5 ml of jugular vein blood was collected from each. Furthermore, Nested-polymerase chain reaction (PCR) targeting the mitochondrial cytochrome b gene of Plasmodium was performed on the same samples using DW2/DW4 primers (primary reaction), followed by NCYBINF/NCYBINR primers (nested reaction). Moreover, risk factors associated with infection, including age, sex, geographic location, season, and environmental factors, were evaluated. Results: No Plasmodium-like parasites or other hemoparasites were observed in any smear. All erythrocytes appeared morphologically normal, with no ring forms, trophozoites, schizonts, or gametocytes detected. Neither the first-round nor the nested amplification yielded any detectable cytochrome b gene of Plasmodium. Conclusion: This study revealed no evidence of Plasmodium infection in dromedary camels in Sistan and Baluchestan province, Iran, as neither microscopic examination nor nested PCR targeting the mitochondrial cytochrome b gene detected any parasite DNA or blood-stage forms.
شیوع و عوامل خطر عفونت پلاسمودیوم در شترهای تک کوهانه (Camelus dromedarius) سیستان و بلوچستان، ایران

چکیده مقدمه: شناخت عفونتهای پلاسمودیوم در شترها برای سلامت حیوانات و ارزیابی خطرات بالقوه انتقال مشترک بین انسان و دام از اهمیت بالایی برخوردار است. از این رو، مطالعه حاضر با هدف بررسی شیوع عفونت پلاسمودیوم در شترها در استان سیستان و بلوچستان ایران انجام شد. مواد و روشها: یک مطالعه مقطعی از نوامبر 2022 تا ژوئیه 2024 انجام شد. در مجموع 270 شتر (111 نمونه در بهار، 93 نمونه در تابستان، 41 نمونه در پاییز و 25 نمونه در زمستان) به طور تصادفی از استان سیستان و بلوچستان انتخاب شدند و 5 میلیلیتر خون ورید وداج از هر کدام جمعآوری شد. علاوه بر این، واکنش زنجیرهای پلیمراز تودرتو (PCR) با هدف قرار دادن ژن سیتوکروم b میتوکندریایی پلاسمودیوم بر روی همان نمونهها با استفاده از آغازگرهای DW2/DW4 (واکنش اولیه) و به دنبال آن آغازگرهای NCYBINF/NCYBINR (واکنش تودرتو) انجام شد. نتایج: هیچ انگل شبه پلاسمودیوم یا سایر انگلهای خونی در هیچ اسمیری مشاهده نشد. همه گلبولهای قرمز از نظر مورفولوژیکی طبیعی به نظر میرسیدند و هیچ شکل حلقهای، تروفوزوئیت، شیزونت یا گامتوسیت مشاهده نشد. نه تکثیر دور اول و نه تکثیر تودرتو هیچ ژن سیتوکروم b قابل تشخیصی از پلاسمودیوم را نشان ندادند. نتیجهگیری: این مطالعه هیچ مدرکی دال بر عفونت پلاسمودیوم در شترهای یک کوهانه در استان سیستان و بلوچستان ایران نشان نداد، زیرا نه بررسی میکروسکوپی و نه PCR تو در تو که ژن سیتوکروم b میتوکندری را هدف قرار میدهد، هیچ DNA انگلی یا اشکال مرحله خونی را شناسایی نکرد.
