چکیده
Introduction: Among the major toxic constituents of snake venoms, phospholipases A₂ (PLA₂) represent a highly diverse enzyme family responsible for a variety of pathological effects, including pronounced myotoxicity and neurotoxicity. Materials & Methods: This study focuses on presynaptically active PLA₂ neurotoxins, β-bungarotoxin (BuTx), taipoxin, crotoxin, notexin, and ammodytoxin C (Amtx). We examined and compared their neuromuscular blocking actions using indirectly stimulated chick biventer cervicis nerve-muscle preparations. Results: BuTx was tested at three concentrations (47, 4.7, and 0.47 nM), producing time and concentration-dependent neuromuscular blockade. It abolished twitch responses within 60 minutes at 47 nM, 120 minutes at 4.7 nM, and 180 minutes at 0.47 nM. All other toxins tested also induced complete neuromuscular paralysis at the given concentrations. Our findings demonstrate that these neurotoxins impair neuromuscular transmission in a dose-dependent manner, ultimately resulting in irreversible muscle paralysis. Among them, BuTx displayed the highest potency, while Amtx was the least potent. To assess postsynaptic function, contracture responses to exogenous acetylcholine (ACh) (1 mM), carbachol (20 μM), and potassium chloride (40 mM) were recorded before and after toxin exposure in the absence of nerve stimulation. These responses remained unchanged, indicating that postsynaptic sensitivity was preserved. Conclusion: A deeper understanding of the mechanisms underlying venom-induced neurotoxicity may improve diagnostic accuracy and support the development of effective treatments for snakebite envenomation, reducing associated morbidity and mortality.
مقایسهٔ اثرات انسداد عصبی–عضلانی نوروتوکسینهای فسفولیپاز A₂ پیشسیناپسی در بافت ایزوله عضلهٔ دو قلوی گردن جوجه

مقدمه: از جمله مهمترین ترکیبات سمی موجود در زهر مارها، فسفولیپازهای A₂ (PLA₂) هستند که با بروز آثار شدید میوتوکسیک و نوروتوکسیک ارتباط دارند. مواد و روش کار: این مطالعه بر روی نوروتوکسینهای پیش سیناپسی (PLA₂) تمرکز دارد؛ ترکیباتی که در بروز مسمومیتهای انسانی نقش مهمی دارند. ما اثرات انسداد عصبی–عضلانی این نوروتوکسین ها را با استفاده از بافت ایزوله عصب–عضلهٔ دو قلوی پس گردنی جوجه، مورد بررسی و مقایسه قرار دادیم. نتایج: BuTx در غلظت های (۴۷، ۴٫۷ و ۰٫۴۷ نانومولار) انسداد عصبی–عضلانی وابسته غلظت ایجاد کرد. این نوروتوکسین پاسخهای انقباضی را در مدت ۶۰ دقیقه در غلظت ۴۷ ، ۱۲۰ دقیقه در غلظت ۴٫۷ و ۱۸۰ دقیقه در غلظت ۰٫۴۷ نانومولار بهطور کامل حذف کرد. سایر نوروتوکسین ها نیز در غلظتهای تعیینشده، فلج کامل عصبی–عضلانی ایجاد نمودند. یافتههای ما نشان میدهد که این نوروتوکسینها انتقال عصبی–عضلانی را بهصورت وابسته به دوز مختل کرده و در نهایت منجر به فلج غیرقابل برگشت عضله میشوند. در میان آنها، BuTx بیشترین توان سمی را از خود نشان داد، در حالی که Amtx کمترین توان را داشت. برای ارزیابی عملکرد پسسیناپسی، پاسخهای انقباضی حاصل از استیلکولین (ACh) (۱ میلیمولار)، کارباکول (۲۰ میکرومولار) و کلرید پتاسیم (۴۰ میلیمولار) پیش و پس از مواجهه با توکسین، در غیاب تحریک عصبی ثبت شد. این پاسخها بدون تغییر باقی ماندند که نشاندهندهٔ حفظ حساسیت پسسیناپسی بود. نتیجه گیری: درک عمیقتر سازوکارهای بنیادین نوروتوکسیسیتهٔ ناشی از زهر مار میتواند دقت تشخیص را بهبود بخشیده و به توسعهٔ درمانهای مؤثر برای مسمومیتهای ناشی از گزش مار کمک کند.
