| Introduction: Newcastle disease (ND) is a highly lethal and contagious disease affecting a diverse range of avian species, caused by the Newcastle disease virus (NDV). This study aims to evaluate the cytotoxic effects of NDV on mouse spleen cells and to assess the activation of specific immune responses against NDV mediated by helper T cells. Material and Methods: Newcastle disease viruses evaluated in this study were collected from broiler farms with disease symptoms in Alborz province. Newcastle disease virus isolates are replicated by inoculation in the allantoic cavity of ten-day old embryonated eggs. Varying concentrations of the virus were introduced to the leukocytes in the first and second groups, while the third group served as a control, receiving no NDV. Following incubation, the gene expression levels were quantified using Real-time RT-PCR, and the cytotoxicity of the virus on the cells was assessed through the MTT assay. Result: The expression levels of the genes gata3, t-bet, interleukin-4, and interferon-gamma did not exhibit significant changes in the group of spleen cells exposed to lower viral concentrations. The evaluation of cytotoxic effects of NDV on spleen cells revealed no cytotoxic impact in the first passage of the NDV treatment group; however, cytotoxic effects were noted during the second passage, although these results were not statistically significant (p<0.05). Conclusion: NDV is capable of activating both cellular and humoral immune responses by stimulating the activation of type 1 and type 2 helper T lymphocytes. Moreover, it exerts influence over the survival and proliferation of leukocytes. | ||

| مقدمه: بیماری نیوکاسل (ND) یک بیماری بسیار کشنده و مسری است که طیف متنوعی از گونههای پرندگان را تحت تأثیر قرار میدهد و توسط ویروس بیماری زا نیوکاسل (NDV) ایجاد میشود. هدف از این مطالعه ارزیابی اثرات سایتوتوکسیک NDV بر سلولهای طحال موش و ارزیابی فعالسازی پاسخهای ایمنی اختصاصی علیه NDV با واسطه سلولهای T کمکی است. مواد و روش کار: ویروس های مورد استفاده در این مطالعه از مرغداری های گوشتی در استان البرز که دارای علائم بیماری بودند و پس از تلقیح به داخل تخم مرغ 10 الی 11 روزه عاری از پاتوژن جداسازی شد. غلظتهای مختلفی از ویروس به لکوسیتهای گروههای اول و دوم اضافه شد، در حالی که گروه سوم به عنوان کنترل در نظر گرفته شد و هیچ ویروسی دریافت نکرد. پس از انکوباسیون، سطح بیان ژن با استفاده از Real-time RT-PCR اندازه گیری شد و اثر سیتوتوکسیک ویروس بر روی سلولها از طریق سنجش MTT ارزیابی شد. نتیجه: سطح بیان ژنهای gata3، t-bet، اینترلوکین-4 و اینترفرون گاما در گروه سلولهای طحال که در معرض غلظتهای پایینتر ویروس قرار گرفته بودند، تغییرات معنیداری نشان نداد. ارزیابی اثرات سیتوتوکسیک NDV بر سلولهای طحال، هیچ اثر سیتوتوکسیک در پاساژ اول گروه تحت درمان با NDV نشان نداد. با این حال، اثرات سیتوتوکسیک در طول پاساژ دوم مشاهده شد، اگرچه این نتایج از نظر آماری معنیدار نبودند (p<0.05). نتیجهگیری: NDV قادر به فعال کردن پاسخهای ایمنی سلولی و هومورال با تحریک فعالسازی لنفوسیتهای T کمکی نوع 1 و نوع 2 است. علاوه بر این، بر بقا و تکثیر لکوسیتها تأثیر میگذارد. | ||
