| Introduction: Brucellosis, also known as Malta fever, is a significant zoonotic infectious disease in Iran, causing considerable public health and economic complications. Human infection is primarily acquired through the consumption of unpasteurized milk and dairy products. Conventional diagnostic approaches, such as culture and serology, are limited by low sensitivity, biosafety risks, and potential cross-reactivity. Molecular methods, particularly real-time polymerase chain reaction (real-time PCR), offer greater sensitivity and specificity. This cross-sectional study, investigated the presence of Brucella spp. in raw milk and traditional dairy products (cream, cheese, and ice cream) collected from local markets in Semnan, Iran. Materials & Methods: A total of 95 samples were analyzed using real-time PCR with SYBR Green dye, targeting the bcsp31 gene for genus-level detection and species-specific primers for Brucella abortus and Brucella melitensis. Additionally, the limit of detection (LOD) was evaluated using the serial dilution of Brucella standard strain. Results: The results revealed that among the samples tested, four samples were positive for Brucella spp. Specifically, three samples were confirmed as B. abortus, and one sample was identified as B. melitensis. Following propagation, DNA fragments of 498 bp and 731 bp, corresponding to B. abortus and B. melitensis, respectively, were successfully detected in the infected samples. Moreover, no significant overall difference in Brucella prevalence was found across products and bacterial load analysis revealed higher median colony-forming units (CFU) in cheese compared to milk and ice cream. Conclusion: These findings highlight the presence of Brucella contamination in dairy products sold through informal channels, emphasizing the need for improved monitoring, strict control strategies, and consumer education to reduce disease transmission. | ||

| ردیابی گونه های سالمونلا در شیرخام و فراورده های لبنی عرضه شده در فروشگاه های سنتی شهر سمنان با روش Real Time PCR | ||

| مقدمه: بروسلوز یا تب مالت یکی از شایعترین بیماریهای عفونی مشترک در ایران می باشد که از دو جنبه بهداشتی و اقتصادی حائز اهمیت است. مصرف شیر و فرآورده های لبنی غیرپاستوریزه یکی از اصلی ترین راه های انتقال آلودگی گونه های باکتری بروسلا به انسان میباشد. با توجه به ویژگی هایی مانند سخت رشد بودن، خطرات مرتبط با کشت، و وجود واکنش های متقاطع با باکتری های دیگر در آزمون سرولوژیک از روشهای با حساسیت و اختصاصیت بالا مانند واکنش زنجیرهای پلیمراز استفاده می شود که از میان انواع PCR، از روش Real-time PCR به عنوان روش بسیار مناسب یاد شده است. اهداف: در این مطالعه مقطعی ، وجود گونههای بروسلا را در شیر خام و محصولات لبنی سنتی (خامه، پنیر و بستنی) جمعآوریشده از بازارهای محلی در سمنان، ایران بررسی کردند.مواد و روش کار: در مجموع 95 نمونه با استفاده از پرایمرهای جنس عمومی و اختصاصی گونههای باکتری بروسلا، ژنهای 31 bcsp، BMEI 1162 و ژن alkB با کمک روش Real-time PCR مورد ارزیابی قرار گرفتند. پس از تکثیر، نمونههای آلوده به باکتری بروسلا آبورتوس حاوی قطعهای به اندازه 498 جفت باز و نمونههای آلوده به بروسلا ملیتنسیس حاوی قطعهای به اندازه 731 جفت باز بودند.همچنین، حد تشخیص (LOD) با استفاده از رقت سریالی سویه استاندارد بروسلا ارزیابی شد. نتایج: نتایج این مطالعه نشان داد که 4 نمونه به باکتری بروسلا آلوده بودند که از این میان 3 نمونه به باکتری بروسلا آبورتوس و 1 نمونه به باکتری بروسلا ملیتنسیس آلودگی داشتند. علاوه بر این، هیچ تفاوت معنیداری در شیوع بروسلا در بین محصولات یافت نشد و تجزیه و تحلیل شمارش باکتریایی، میانگین CFU بالاتری را در پنیر در مقایسه با شیر و بستنی نشان داد. نتیجه گیری: بنابراین با توجه به آلودگی شیر و فراوردههای آن به باکتری بروسلا و با عنایت به زیانهای بهداشتی و اقتصادی این بیماری، لازم است برنامههای کنترلی و نظارتی دقیق تر و کارآمد تری جهت پیشگیری و کنترل بیماری اتخاذ گردد. | ||
