| Introduction: This study comprehensively evaluated the dose- and time-dependent effects of di-(2-ethylhexyl) phthalate (DEHP), a widely used environmental contaminant, on the spleen of Sparidentex hasta over a 14-day exposure period. Materials & Methods: Fish were exposed to five concentrations of DEHP (0, 5, 10, 50, and 100 µg/g), and splenic responses were assessed through quantitative morphometric indices, including spleen weight (SW), spleen somatic index (SSI), and spleen-to-length ratio (SLR), as well as qualitative histopathological analysis. Statistical comparisons were performed using SPSS 26, and graphical representations were generated in Excel. Results: Results demonstrated significant dose- and time-related increases in SW (0.10 g in controls vs 0.27 g at 100 µg/g, P<0.05), SSI (0.089% vs 0.276%, P<0.05), and SLR (0.563% vs 1.698%, P<0.05). Histopathological examinations revealed progressive structural alterations with increasing DEHP concentrations, including sinusoidal congestion, lymphocytic hyperplasia, melanomacrophage center (MMC) accumulation, and focal necrosis. Lower doses induced mild adaptive responses, whereas higher exposures resulted in pronounced tissue disorganization and immune activation. Strong correlations between morphometric indices and microscopic lesions suggested that splenic enlargement reflects both oxidative stress and immunotoxic effects. Conclusion: These findings underscore the sensitivity of the spleen as a reliable biomarker for detecting sublethal immunotoxicity in fish exposed to environmental pollutants. Overall, this study highlights the importance of integrating quantitative indices with histopathological evaluation to assess the ecotoxicological impacts of DEHP, providing valuable insights for environmental monitoring, fish health management, and aquaculture sustainability. The results further emphasize the potential ecological and economic consequences of DEHP contamination in aquatic ecosystems. | ||

| این پژوهش بهصورت جامع اثرات وابسته به دوز و زمان دی-(۲-اتیلهگزیل) فتالات (DEHP) ـ یکی از آلایندههای رایج محیطزیستی ـ را بر طحال ماهی Sparidentex hasta طی یک دورهی ۱۴ روزه بررسی کرد. ماهیان در پنج غلظت مختلف DEHP (۰، ۵، ۱۰، ۵۰ و ۱۰۰ میکروگرم بر گرم) تیمار شدند و پاسخهای طحالی از طریق شاخصهای مورفومتریک کمی شامل وزن طحال (SW)، شاخص طحالتنی (SSI) و نسبت طحال به طول بدن (SLR) به همراه تحلیلهای بافتشناسی کیفی ارزیابی گردید. آزمونهای آماری با نرمافزار SPSS نسخه ۲۶ انجام و نمودارها با نرمافزار Excel ترسیم شدند. نتایج نشان داد افزایش معنیدار وابسته به دوز و زمان در مقادیر SW (۰٫۱۰ گرم در گروه شاهد در مقابل ۰٫۲۷ گرم در غلظت ۱۰۰ میکروگرم بر گرم؛ p<0.05)، SSI (۰٫۰۸۹٪ در مقابل ۰٫۲۷۶٪؛ p<0.05) و SLR (۰٫۵۶۳٪ در مقابل ۱٫۶۹۸٪؛ p<0.05) رخ داد. بررسیهای بافتشناسی نشان داد که با افزایش غلظت DEHP، تغییرات ساختاری بهتدریج تشدید میشود و شامل احتقان سینوزوئیدی، هیپرپلازی لنفوسیتی، تجمع مراکز ملانومکروفاژی (MMC) و نکروز موضعی است. دوزهای پایینتر موجب پاسخهای سازشی خفیف شدند، در حالیکه غلظتهای بالاتر منجر به بینظمی بافتی شدید و فعالشدن پاسخهای ایمنی گردید. همبستگی قوی میان شاخصهای مورفومتریک و ضایعات میکروسکوپی نشان داد که افزایش اندازه طحال بازتابی از استرس اکسیداتیو و سمیت ایمنی است. این یافتهها حساسیت طحال را بهعنوان یک نشانگر زیستی معتبر برای ارزیابی اثرات ایمنوتوکسیک DEHP در ماهیان تأیید میکند. در مجموع، نتایج این پژوهش بر اهمیت تلفیق شاخصهای کمی و تحلیلهای بافتشناسی برای پایش اکوتوکسیکولوژیک تأکید دارد و پیامدهای بالقوه آلودگی DEHP بر سلامت ماهی، پایداری آبزیپروری و ایمنی غذایی را برجسته میسازد. | ||
