Application of Cryotherapy in the Treatment of Cutaneous Leishmaniasis in BALB/c Mice Compared with Commercial Injectable Treatments | ||

| Abstract Cutaneous leishmaniasis (CL), caused by Leishmania species, is a parasitic disease prevalent in tropical and subtropical regions. Currently, pentavalent antimonial compounds such as pentostam (sodium stibogluconate) and glucantime (meglumine antimoniate) are considered first-line treatments for CL. In this study the efficacy of cryotherapy administered alone and in combination with commercial drugs for treating CL compared to monotherapy with commercial treatments (glucantime and amphotericin B) in BALB/c mice was investigated. Mice infected with Leishmania major were divided into six groups (n=5 per group). Following lesion development, glucantime was administered daily via direct intralesional injection, while amphotericin B was administered intraperitoneally as a single daily dose for three weeks. Cryotherapy was performed six times over a maximum period of three weeks. At the conclusion of the treatment period, two mice from each group were euthanized under sterile conditions, and their spleens were harvested to determine parasite load through culture in RPMI 1640 medium. The results demonstrated a significant reduction in lesion size and accelerated wound healing in groups treated with cryotherapy combined with glucantime or amphotericin B compared to the control group. In the cryotherapy-glucantime group, complete wound closure was achieved by the third week, with no residual nodules or lesion expansion, as observed in the untreated control group. Notably, at the end of the treatment period, no parasites were detected in the spleens of any treatment group. This study supports cryotherapy as an effective adjunctive strategy for enhancing the efficacy of conventional drugs in cutaneous leishmaniasis. | چکیده لیشمانیوز جلدی (CL)، که توسط گونههای لیشمانیا ایجاد میشود، یک بیماری انگلی شایع در مناطق گرمسیری و نیمهگرمسیری است. در حال حاضر، ترکیبات آنتیموان پنج ظرفیتی مانند پنتوستام (سدیم استیبوگلوکونات) و گلوکانتیم (مگلومین آنتیموان) به عنوان درمانهای خط اول برای لیشمانیوز جلدی در نظر گرفته میشوند. در این مطالعه، اثربخشی کرایوتراپی به تنهایی و در ترکیب با داروهای تجاری برای درمان لیشمانیوز جلدی در مقایسه با مونوتراپی با درمانهای تجاری (گلوکانتیم و آمفوتریسین B) در موشهای BALB/c بررسی شد. موشهای آلوده به لیشمانیا ماژور به شش گروه (n=5 در هر گروه) تقسیم شدند. پس از ایجاد ضایعه، گلوکانتیم روزانه از طریق تزریق مستقیم داخل ضایعه تجویز شد، در حالی که آمفوتریسین B به صورت داخل صفاقی به صورت یک دوز روزانه به مدت سه هفته تجویز شد. کرایوتراپی شش بار در حداکثر دوره سه هفته انجام شد. در پایان دوره درمان، دو موش از هر گروه در شرایط استریل کشته شدند و طحال آنها برای تعیین میزان انگل از طریق کشت در محیط کشت RPMI 1640 برداشت شد. نتایج نشان دهنده کاهش قابل توجه اندازه ضایعه و تسریع بهبود زخم در گروههای تحت درمان با کرایوتراپی همراه با گلوکانتیم یا آمفوتریسین B در مقایسه با گروه کنترل بود. در گروه کرایوتراپی-گلوکانتیم، بسته شدن کامل زخم تا هفته سوم حاصل شد، بدون هیچ ندول باقی مانده یا گسترش ضایعه، همانطور که در گروه کنترل درمان نشده مشاهده شد. نکته قابل توجه این است که در پایان دوره درمان، هیچ انگلی در طحال هیچ یک از گروههای درمانی مشاهده نشد. این مطالعه از کرایوتراپی به عنوان یک استراتژی کمکی موثر برای افزایش اثربخشی داروهای مرسوم در لیشمانیوز جلدی پشتیبانی میکند. | ||
