چکیده
Cystic echinococcosis is a significant zoonotic disease, transmitted primarily from dogs to intermediary hosts such as humans, sheep, and cattle. This disease is caused by the larval stage of the Echinococcus granulosus tapeworm. Cystic echinococcosis can lead to severe health complications, including liver and lung cysts, which can cause organ dysfunction and even death if left untreated.
Within this study, serum specimens were collected from 25 newborn babies as the negative control group, while an additional 25 serum samples were gathered from surgically confirmed cases of CE to serve as the positive control group. In this study, an Iranian native antigen B was used to design the specific detection of hydatidosis in humans using an AuNP-ELISA method. The AgB isolated from sheep CE fluid was used for the design using the AgB-ELISA method. This study was performed in three stages. At first used the commercial ELISA kit, the second method used native AgB to design the ELISA method, and at last, were used gold nanoparticles (AuNPs) with anti-human conjugate combination and AgB-ELISA method.
According to the results, sensitivity and specificity for the diagnosis of hydatidosis in commercial ELISA kit was 92% and 100% respectively, in AgB-ELISA method 96% and 100%, and in Gold Nanoparticle-ELISA method 100% and 100% respectively.
Using Ag B in ELISA design for hydatidosis diagnosis has attracted researchers in recent years. The results have shown that using AgB in ELISA design is so valuable, but the sensitivity will be increased significantly if used gold nanoparticles with anti-human conjugate, especially in lower titer which needs accuracy.

ارزیابی کارآیی تشخیصی تکنیک توسعه یافته استفاده از نانوذرات طلا در تشخیص اکینوکوکوزیس به روش ELISA

بیماری کیست هیداتید (اکینوکوکوزیس سیستیک) یک بیماری زئونوتیک مهم است که عمدتاً از سگ‌ها به میزبان‌های واسطه‌ای مانند انسان، گوسفند و گاو منتقل می‌شود. این بیماری توسط مرحله لاروی کرم نواری اکینوکوکوس گرانولوزوس ایجاد می‌شود. کیست هیداتید می‌تواند باعث ایجاد عوارض جدی سلامتی شود، از جمله کیست‌های کبد و ریه که می‌توانند باعث اختلال در عملکرد اعضا و حتی مرگ در صورت عدم درمان شوند.
در این مطالعه، نمونه‌های سرمی از ۲۵ نوزاد تازه متولد شده به عنوان گروه کنترل منفی جمع‌آوری شد، و ۲۵ نمونه سرمی دیگر از موارد جراحی تأیید شده اکینوکوکوزیس سیستیک به عنوان گروه کنترل مثبت جمع‌آوری شد. در این مطالعه، از آنتی‌ژن B بومی ایران برای شناسایی خاص هیداتیدوزیس در انسان‌ها از طریق روش AuNP-ELISA استفاده شد. آنتی‌ژن B استخراج شده از مایع کیست هیداتید گوسفند برای طراحی با استفاده از روش AgB-ELISA استفاده شد. این مطالعه در سه مرحله انجام شد. اولین مرحله استفاده از کیت تجاری ELISA بود؛ درمرحله دوم از آنتی‌ژن B بومی برای طراحی روش ELISA استفاده شد؛ و مرحله آخر استفاده از نانوذرات طلا (AuNPs) با روش AgB-ELISA بود.
بر اساس نتایج، حساسیت و ویژگی برای تشخیص هیداتیدوزیس در کیت تجاری ELISA به ترتیب ۹۲٪ و ۱۰۰٪ بود؛ در روش AgB-ELISA به ترتیب ۹۶٪ و ۱۰۰٪، و در روش نانوذرات طلا-ELISA به ترتیب ۱۰۰٪ و ۱۰۰٪ بود.
استفاده از آنتی‌ژن B در طراحی ELISA برای تشخیص هیداتیدوزیس اخیراً توجه پژوهشگران را جلب کرده است. نتایج نشان داده است که استفاده از آنتی‌ژن B در طراحی ELISA بسیار ارزشمند است. با این حال، استفاده از روش تشخیصی با استفاده از نانو ذرات طلا و کونژوگه انسانی دارای حساسیت بیشتریدر تشخیص می باشد.
