چکیده
Fowl Adenoviruses (FAdVs) are widely distributed pathogens across the globe. The FAdVs from serotypes FAdV 2, 3, 8a, 8b, 9, and 11 are responsible for inclusion body hepatitis (IBH). Recently, increased mortality and IBH-suspected lesions were observed in 8-10-day-old broiler chickens in West Azerbaijan Province, Iran. In this regard, the present study aimed to compare penton and hexon genes of ADDV11 in the molecular detection of IBH in broiler chickens. In total, 100 liver specimens were collected from 10 suspected farms, and their DNAs were extracted. Two polymerase chain reactions (PCRs) were applied; one targeting the L1 region of the hexon gene and another aiming at the penton gene. Based on the findings, 60% of samples showed positive results in both PCRs and phylogenetic analysis clustered the studied viruses into serotype 11 (species D) FAdV. The detected FAdVs also shared a multitude of homologies with previously published serotype 11 viruses from Iran and those identified in Pakistan, Saudi Arabia, India, China, and Canada. This research not only provides an update on circulating FAdVs in Iran, but also introduces the penton gene as an alternative target for IBH diagnosis. Considering that IBH is a primary disease in Iran with both horizontal and vertical routes of transmission, urgent preventive measures are needed.

مطالعه مقایسه ای ژنهای پنتون و هگزان سروتیپ 11 آدنوویروس مرغی در تشخیص مولکولی بیماری هپاتیت انکلوژن بادی در طیور گوشتی

آدنوویروس های طیور (FAdVs) پاتوژن هایی هستند که به طور گسترده در سراسر جهان توزیع شده است.سروتیپ های آدنوویروس های طیور شامل 2، 3، 9,8a,8b و 11 مسئول هپاتیت دارای گنجیدگی(IBH) هستند. اخیرا، افزایش مرگ و میر و ظایعات IBH در جوجه های گوشتی هشت تا ده روزگی در استان آذربایجان غربی ایران مشاهده شد. مطالعه حاظر به منظور مقایسه ژنهای پنتون و هگزان آدنوویروس ماکیان در تشخیص مولکولی بیماری هپاتیت انکلوژن بادی در طیور گوشتی انجام شد. 100 نمونه کبد از ده مزرعه مشکوک جمع آوری شد و از آنها DNA استخراج شد. دو PCR در این مطالعه انجام شد، یکی ناحیه L1 ژن هگزون و دیگری ژن پنتون را هدف قرار می دهد. نتایج: 60 درصد نمونه ها نتایج مثبتی در هر دو PCR نشان دادند و آنالیز فیلوژنتیک ویروس های ما را در سرو تیپ 11 ( گونه D) آدنوویروس های طیور دسته بندی کرد. آدنوویروس های طیور شناسایی شده نیز با ویروس های سروتیپ 11 منتشر شده قبلی از ایران و ویروس هایی که در پاکستان، عربستان سعودی، هند، چین و کانادا شناسایی شده اند مشابهت بالایی داشتند. این مطالعه نه تنها به روز رسانی در مورد آدنوویروس های طیور در گردش در ایران را ارائه می کند بلکه ژن پنتون را به عنوان یک هدف جایگزین برای تشخیص IBH معرفی می کند. با توجّه به اینکه IBH به عنوان یک بیماری اولیه در ایران و راههای انتقال ان به صورت افقی و عمودی، اقدامات پیشگیرانه عاجل ضروری می باشد.
