| چکیده | ||
| The genus Sedlitzia (Amaranthaceae) includes four herbaceous and one shrubby species. Sedlitzia rosmarinus is resistant to salinity that plays an important role in soil conservation. In this research, RAPD molecular markers were used to evaluate the genetic diversity among 85 plant samples from 17 populations of S. rosmarinus. Five primers were used to perform RAPD reactions and a total of 400 DNA bands were obtained. After forming 0 and 1 matrix with the help of GenAlex 6.41 and UPGMA methods using SHAN clustering model, NTSYS Ver. 2.02 software was used to interpret RAPD analysis data. Molecular analysis of variance (AMOVA) was performed to estimate genetic diversity out of 85 individuals. Out of the total diversity, it was calculated that, the highest amount of genetic diversity i.e. 69% was related to intra-population and 31% to inter-population diversity, respectively. | ||

| جنس Seidlitzia (تاجخروسیان) دارای سه گونه علفی و یک گونه درختچهای است. گیاه اشنان (Seidlitzia rosmarinus) یکی از گیاهان مقاوم جوامع شورپسند است که نقش مهمی در حفاظت خاک ایفا میکند. در این تحقیق، نشانگر مولکولی RAPD برای بررسی تنوع ژنتیکی جمعیت گونه مذکور، 85 نمونه گیاهی از 17 جمعیت، مورد مطالعه قرار گرفت. تعداد پنج آغازگر تصادفی در انجام واکنش RAPD به کار گرفته شد و در کل 400 باند DNA به دست آمد. بیشترین تعداد باندها از دو آغازگر OPM-06 وOPS-07 به دست آمد. در تجزیه کلاستر ژنوتیپها برای تفسیر دادههای آنالیز RAPD از نرمافزار NTSYS Ver. 2.02 استفاده شد. ماتریس تشابه ایجاد شده در نرمافزار NTSYS با روش UPGMA با استفاده از مدل خوشهبندی SAHN به دست آمد. در تحقیق حاضر، جمعیت کاشان (PSE) با بیشترین فاصله، به عنوان گروه خواهری سایر جمعیتهای موجود در نظر گرفته شد. بعد از تشکیل ماتریس صفر و یک با کمک نرمافزار Gen Alex 6.41، برای تفسیر دادههای آنالیز RAPD، از نرمافزار آنالیز واریانس مولکولی (AMOVA) به منظور برآورد تنوع ژنتیکی استفاده شد. از مجموع کل تنوع، بیشترین میزان تنوع ژنتیکی یعنی 69% آن مربوط به تنوع درون جمعیتی، 31% تنوع بین جمعیتی محاسبه شد. با توجه به اهداف این تحقیق، میتوان نتیجه گرفت که نشانگر مولکولی RAPD روشی سودمند برای بررسی پراکندگی ژنتیکی جمعیت اشنان بوده است. | ||
