شناسایی مولکولی سویههای بروسلا آبورتوس در ایران با استفاده از روشهای جدید تغییر یافته واکنش زنجیرهای پلیمراز و Real-time PCR | ||

| Abstract | ||
| بروسلوز در درجه اول یک بیماری زئونوز پراهمیت در سراسر جهان است که توسط گونههای بروسلا ایجاد میشود. جنس بروسلا دارای گونههای بسیار عفونی است که به عنوان عوامل تهدید کننده زیستی طبقه بندی میشوند. شناسایی بروسلا یک فرآیند زمان گیر و سخت است که میتواند پرسنل را در معرض خطر عفونتهای آزمایشگاهی قرار دهد. این تحقیق با هدف ارائه یک روش آزمایش جدید برای واکنش زنجیرهای پلیمراز معمول و واکنش زنجیرهای پلیمراز زمان واقعی (real-time PCR) برای شناسایی گونههای بروسلا آبورتوس انجام شده است. در این مطالعه دو آغازگر (bru ab2) بر اساس جایگاههای منحصر به فرد کد کننده انتقال دهنده خودی ((autotransporter واجد تکرارهای رشته بتا پروتئین (شماره دسترسی YP00113760) طراحی شده است. پنجاه و شش سویه بروسلا (جدایههای مرجع، واکسن و جداسازی شده از مزرعه) و Yersiniaenterocolitica به عنوان یک جدایه غیر بروسلا در سیستم معمول و واکنش زنجیرهای پلیمراز زمان واقعی مورد ارزیابی قرار گرفتند. نتایج نشان داد که با مقادیر ng 0.4 وfg 400 DNA ژنومی سویههای بروسلا آبورتوس میتوانند با استفاده از روش معمول و واکنش زنجیرهای پلیمراز زمان واقعی شناسایی شوند. پرایمرهایbru ab2 برای هر دو روش واکنش زنجیرهای پلیمراز مناسب بودند. هر دو روش برای شناسایی تمام سویههای باکتری اختصاصی بوده اما آزمایش واکنش زنجیرهای پلیمراز در زمان واقعی 1000 برابر حساستر از روش واکنش زنجیرهای پلیمراز معمولی بود. از این رو این روش به عنوان روش انتخابی قدرتمند جهت شناسایی تمام گونههای بروسلا آبورتوس پیشنهاد میشود. | ||
