| The study was intended to investigate the morphological, morphometric and molecular characterization of eight geographical isolates of the entomopathogenic nematode Steinernema feltiae from northwest of Iran. The results showed no detectable morphological variations, but significant morphometric variations were recorded among the isolates. Clustering based on morphometry of infective juveniles, classified the isolates in the following three separate groups: group I consists of IRA30 and IRA25; group II includes IRA21, IRA23, IRA34, IRA28 and IRA21 and group III contains of IRA17. Clustering the isolates based on the male's morphometric characters, yielded nearly similar results and four groups were constructed as follows: group I is made up of IRA22, IRA23 and IRA21; group II holds IRA25, IRA34 and IRA28; group III contains IRA30 and IRA17 constitutes group IV. No intraspecific variation was observed among PCR-RFLP patterns of the native isolates resulted from16 restriction enzymes but significant variations were recorded within the isolate Pumping by HinfI, MspI and MboI. Although these markers serve as effective tools in separating the native isolates from the exotic ones, they are unable to detect variations among native isolates. Therefore, only ITS-rDNA sequence based phylogenetic analysis can effectively clarify the intraspecific variations of S. feltiae. | ||

| با هدف مطالعه صفات ریختشناختی، ریختسنجی و مولکولی جدایههای نماتود بیمارگر حشرات Steinernema feltiae، هشت جدایه از شمالغرب ایران مورد شناسایی و بررسی قرارگرفت. در بین جدایهها تنوع ریختشناختی مشاهده نگردید ولی تنوع ریختسنجی قابل توجهی ردیابی و جدایههای جغرافیایی بر اساس صفات لاروهای آلودهکننده در سه گروه متفاوت قرار گرفتند، گروه اول متشکل از جدایههای IRA25 و IRA30، گروه دوم شامل IRA21، IRA28، IRA34،IRA22 و IRA23 و IRA17 با بیشترین فاصله گروه سوم را تشکیل داد. گروهبندی بر اساس دادههای ریختسنجی افراد نر، جدایهها را در چهار گروه قرار داد. جدایههای IRA22 ، IRA23 و IRA21 یک گروه، IRA28، IRA34 و IRA25 گروه دوم و IRA30 و IRA17 هر کدام در یک گروه جداگانه قرارگرفتند. مشابه گروهبندی حاصل از لاروهای آلوده کننده، جدایه IRA17 به صورت جداگانه و با بیشترین فاصله نسبت به بقیه گروهها قرارگرفت. در بررسی الگوی PCR-RFLP ناحیه ITS-rDNA با شانزده آنزیم برشی اختلافی بین جدایهها مشاهده نگردید ولی بین جدایههای ایرانی و جدایه Pumping اختلاف قابل ملاحظهای به عنوان تنوع درون گونهای مشاهده گردید. نتایج نشان داد هر چند که الگوی PCR-RFLP ناحیه ITS-rDNA آنزیمهای HinfI، MspI و MboI جهت تفکیک جدایههای مورد بررسیحداقل از جدایه Pumping نشانگر کارآمدی میباشد اما در تفکیک جدایههای بومی فاقد کارایی بوده و لازم است روشهای کارآمدتر، از جمله تجزیه و تحلیل نسب شناختی مبتنی بر ترادف نوکلئوتیدی ITS-rDNA یا دیگر نواحی ژنومی مناسب، مورد ارزیابی و استفاده قرار گیرد. | ||
